Neural function and structure

Electrical imaging data illuminates cell health and morphology, while non-invasive longitudinal electrophysiological measurements reveal neural network formation. By combining electrical imaging with electrophysiology, Pixel enables you to simultaneously track development of neuronal activity, cell health, morphology, and viability in the same culture.

CAR-T cell therapy in spheroids

How do CAR-T cells attack tumors in real-time? Pixel electrical imaging captures the dynamic interactions between T cells and tumor spheroids, revealing both structural changes and immune cell activity.

In this experiment, HeLa-CD19 spheroids were incubated with CD19-targeting T cells from ProMab Biotechnologies for 72 hours. The left panel shows an untreated spheroid growing over time, while the right demonstrates CAR-T cells reducing the tumor mass within 24 hours after their addition. Pixel enables visualization of T cell movement as well as their impact on the tumor microenvironment.

CAR-T cell therapy in 2D cell cultures

Sensitive detection of cell death in physiologically relevant in vitro models is critical to improve CAR-T cell therapies. With single-cell resolution, Pixel electrical imaging allows you to closely monitor interactions between T and tumor cells, including changes in cell morphology before death. Here, dying cells with altered morphologies are visible in green and blue.

In this experiment, HeLa-CD19 cells were incubated with CD19-targeting T cells from ProMab Biotechnologies for 72 hours. The left panel shows untreated cells, while the middle and right panels are seeded with 1:6 and 1:10 CAR-T:tumor cells at 24 hours, respectively. Cell death is visible even at low CAR-T: tumor cell ratios.

Neuronal cells

How do neuronal cells interact in co-cultures? Neurons, astrocytes, and microglia have unique phenotypes and live-cell behavior that can be identified by electrical imaging. Pixel allows you to track the behavior and response of each cell type in mono or co-cultures, like these neuronal cells from BrainXell.

ALS in motor neurons

How does ALS alter the structure and behavior of motor neurons? By applying Pixel high-dimensional electrical imaging to Axol Bioscience’s axoCells™, we can identify different phenotypes in iPSC-derived motor neurons from both healthy donors and ALS patients.

These videos show phenotypic differences between healthy patient cells and those from ALS patients with two different genetic mutations, using 3 of Pixel’s 20+ morphological and functional parameters.

Endothelial cell motility and tissue barrier

Pixel electrical imaging lets you observe a variety of properties of endothelial cells, such as motility (left) and tissue barrier formation (right). Pixel’s high sensitivity and spatial accuracy allows you to monitor the formation of cell junctions and tissue barrier (right, deep blue) in real time. With Pixel 96- and 384-well microplates, you can also test the barrier’s response to a range of molecules.
